I have applied for leave without pay for 2 years to pursue a regular course. But my department rejected it without giving any reason. Can I sue my department for not giving me leave.

A.I guess you are working in a public sector organization. If they have policy of sabbatical then must give you a reason for rejection if not then you will have take it up with the HR and find out a tailored solution for this scenario. Filing a case against the organization is possible but one has to look into many factors.
